      Abstract:There are two fertility restorer genes,Rf5 and Rf6 in 9311 and two sterility genes orfH79 and orf216 in YTA.To study the interaction between two restorer genes and two CMS genes in Honglian (HL) rice,HL-CMS line YTA was crossed with the restore line 9311.Then,F1 was backcrossed repeatedly as the male parent with the YTA as recurrent male parent for more than eight generations.Based on the sequences of Rf5 ,we designed the specific primers and tested the restorer gene in L-Rf5.Combing with the pollen fertility,we did not find genetic divergence in self-crossed progenies.It suggested that the homozygous L-Rf5 was constructed successfully.By analyzing the expression level of HL-CMS gene orfH79 and orf216 in L-Rf5,we found that the expression level of orfH79 in L-Rf5 decreased distinctly than that in YTA.There is no obvious difference between the homozygous L-Rf5 and the heterozygous L-Rf5.However,The expression of orf216 in L-Rf5 was not significantly decreased compared with YTA.Meanwhile,the Rf1B sequences of YTA,9311,near isogenic lines were cloned,and we found no difference in these materials.This study laid the foundation for molecular analysis of the restoration of fertility gene Rf1B.

    • Ectopic expression,regulation and function of CBF1 genes from Solanum lycopersicoides and Lycopersicon habrochaites in tobacco

      Abstract:The CBF1 gene from Solanum lycopersicoides and Lycopersicon habrochaites was transferred into tobacco(Nicotiana tobacum L.) by agrobacterium-mediated leaf disc method. The transcriptomic differences of transgenic tobacco under low temperature stress were studied. The results showed that under low temperature stress,the low temperature tolerance of transgenic tobacco was higher than that of the wild type when the transgenic tobacco was under low temperature stress. Both SlCBF1 and LhCBF1 genes were overexpressed in ordinary tobacco. There were 53 differentially expressed genes,33 up-regulated genes and 20 down-regulated genes in transgenic tobacco plants expressing SlCBF1 and wild-type tobacco. There were 106 differentially expressed genes,62 genes up-regulated and 44 down-regulated genes in transgenic tobacco plants expressing LhCBF1 gene and wild-type tobacco.It is indicated that SlCBF1and LhCBF1 may play an important role in transgenic tobacco to obtain anti-low temperature. 

      Abstract:The relative water content (RWC), superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), soluble protein, malondialdehyde (MDA) and proline (Pro) under drought stress at 20% PEG 6000 concentration were studied to explore the effect of drought stress on carnation seedlings and evaluate the drought resistance of 9 varieties of carnation.Based on the drought resistance index of these seven physical and chemical indexes, the principal component analysis and comprehensive evaluation of all varieties of carnation were carried out.The results showed that there were significant differences among the indexes of 9 carnation cultivars under water stress (P<0.05).Seven drought resistance indexes were classified into four principal components by principal component analysis, with the cumulative contribution rate of 84.46%. The drought resistance evaluated by the membership function method is in the order of ‘Master’, ‘Tess’, ‘Royal’, ‘Violet’, ‘Flame’, ‘Pink Love’, ‘Mallow Titanium Alloy’, ‘Pink Superior’, ‘Freedom’.

    • Relationship between landscape components and leisure satisfaction of roadside green space in Hefei City

      Abstract:10 roadside green spaces within the second ring road in Hefei City were surveyed with statistical,ecological and psychological methods.The relationship between green space landscape components and recreation satisfaction was explored with questionnaire survey of recreation satisfaction.The results showed that the comprehensive satisfaction of recreation in Hefei was different from each roadside greenbelt,with green space satisfaction scores ranging from 3.044 to 4.320.The overall satisfaction was common.The Rui Garden built in 2010 scored the best,and the East Changjiang road and Nanling road cross garden built in 2000 scored the lowest.By setting up model for landscape components and recreation satisfaction,the green space with high citizen satisfaction had the performance with suitable ratio of green space and pavement surface,appropriate proportion of evergreen and deciduous trees,adequate shade and reasonable width of sidewalks. Results of analyzing the curve estimation regression with SPSS 23.0 analysis software showed that there was a certain curve relationship between the green space landscape components and recreation satisfaction.The corresponding optimal estimation models were established.

    • Detection of main viral species of vegetable virus diseases in Hubei Province

      Abstract:Nine hundred and sixty-six suspected vegetable samples were collected from seven regions in Hubei Province.Seven viruses including Cucumber mosaic virus (CMV),Tobacco mosaic virus (TMV),Tomato spotted wilt virus (TSWV),Tomato mosaic virus (ToMV),Turnip mosaic virus (TuMV),Broad bean wilt virus (BBWV) and Cucumber green mottle mosaic virus (CGMMV) were detected by Dot-ELISA and RT-PCR.The results showed that 695 of the 966 samples were positive for virus infection,accounting for an incidence of 71.9%.Of which,TuMV ranked the highest with an incidence of 86.4%,followed by CMV with an incidence of 26.7%,in brassicaceous vegetables.CMV infected solanaceous,cucurbitaceous and leguminous vegetables with an incidence of 34.3%,59.2% and 56.2%,respectively.CMV,which infected almost all vegetable plants,was the major virus infecting vegetables in Hubei province.For the six viruses detected,they infected plants in six mixed patterns,including TuMV+CMV,TuMV+BBWV,TuMV+CMV+BBWV,CMV+BBWV,CMV+ToMV and CMV+BBWV+TSWV+TMV.Of which,TuMV+CMV and CMV+BBWV were the major combination of the mixed infection in brassicaceous vegetables and the other vegetables,accounting for an incidence of 23.8% and 28.0%,respectively.

    • Phylogenetic analysis of exotic Radopholus similis in China based on rDNA-ITS sequences

      Abstract:To demonstrate genetic divergence of different populations of exotic Radopholus simils in China,analyze their phylogenetic relationship,and provide scientific proof for the origin of these populations,the published universal primers were used to amplify the rDNA-ITS sequences of progeny of single and 25 females,respectively,from 20 populations of R. similis. The amplified products were sequenced,blasted and aligned with public submitted sequences and phylogenetic tree was then constructed. The rDNA-ITS sequenced was 705-713 bp in length,and showed high similarity (≥95%) among different populations,with a divergence of 0-7.5%. The rDNA- ITS1 and ITS2 were 273-276 bp and 151-152 bp in length,with sequences similarity ≥98.58% and ≥98.35%,respectively. The sequences alignment results showed that the ITS sequences had a very high similarity between the progeny of single female and multiple females in the same population,showing 100% sequence identity in 8 populations and ≥97% similarity in the other populations. In addition,the phylogenetic tree with Pratylenchus coffeae as outgroup showed that all R. similis populations were clustered together,but with 4 small branches. Among them, Anthurium andraeanum population RsW was clustered in a single branch,Philodendron sp. population RsH had a longer genetic distance with other members in the same branch,but Calathea zebrine population RsP and C. makoyana RsS had a shorter genetic distance. The results indicated that the R. similis populations in China mainly came from the European Marantaceae and Araceae ornamentals,and the primary origin for the former was banana in Sudan and was complicated for the latter.

      Abstract:Small non-coding RNAs (sRNAs) are a large number of regulators and are significantly important in physiological processes in bacteria.However,few approaches are available for identifying their cellular targets in vivo.In this study,a two-plasmid system based on the initial vectors pMycVec2 and pMycVec1 was established to screen the targets of mycobacterial sRNA.The vector (pMycVec2-D) for cloning sRNA was derived from the vector pMycVec2 and contained the strong promoter rrnBp and an effective transcription terminator,which has been modified to enable tight regulation of the expression of sRNA.The vectors for cloning target genes,pMycVec1-lacZ and pMycVec1-GFP,were constructed from the pMycVec1,which stabilized the transcription with a weak promoter Pwk,and the translation level of target was detected through the reporter genes.The two-plasmid system was validated by detecting the interaction between sRNA and its regulatory target,MicC/ompC mRNA and MicF/ompF mRNA respectively.

    • Effects of a TetR family transcription factor Ms2583 on isoniazid sensitivity in Mycobacterium smegmatis

      Abstract:Bacteria use transcriptional regulation to respond to environmental stresses. However,only a few transcription factors regulating drug resistance have been reported. In this study,a transcriptional factor library of Mycobacterium smegmatis was screened on drug plates. A novel transcriptional factor,encoded by the MSMEG_2583 (Ms2583) gene directly affecting M. smegmatis Isoniazid (INH) sensitivity was characterized. Over-expressing Ms2583 caused a significant increase in INH sensitivity of the transformed cells. Results of analyzing structural characteristics of Ms2583 showed that Ms2583 is a TetR family transcription factor. The bacterial one-hybrid assays and electrophoretic mobility shift assays demonstrated that Ms2583 could directly bind to its own promoter with specific interaction.These findings will provide a clue for studying the regulatory mechanism of TetR family transcription factor Ms2583.

      Abstract:Mycobacterium tuberculosis is the causative agent of tuberculosis (TB), one of the deadliest diseases worldwide. The emergence of multidrug-resistant (MDR) TB and extensive drug-resistant (XDR) TB is a large challenge to TB treatment. In this study, a transcriptional factor library of Mycobacterium smegmatis as a model strain for pathogenic M. tuberculosis was screened on plates containing 15 μg/mL Isoniazid (INH).A novel transcriptional factor encoded by MSMEG_3469 (Ms3469) directly affecting M. smegmatis INH sensitity was characterized. Over-expression of Ms3469 caused a significant increase in INH resistance of M. smegmatis. Results of bioinformatics analyses showed that Ms3469 is a member of the TetR/AcrR family of transcription regulators and forms a homodimer similar to SO_TetR.The electrophoretic mobility shift assays demonstrated that Ms3469 could bind to its own promoter.The direct and specific interaction was further confirmed by bacterial one-hybrid assays.It will provide a clue for studying the regulatory mechanism of bacterial drug resistance in M. smegmatis.

    • Synthesizing FeMoO4 hollow microspheres with gas template method and its photocatalytic reduction of Cr (Ⅵ)

      Abstract:FeMoO4 hollow microspheres as eco-friendly materials with the visible-light activity were prepared from FeNO3 and (NH4)6Mo7O24 through a solvent-thermal method using ethylene glycol and HNO3  mixture as solvents.Crystal structures,compositions and morphologies of the materials obtained were characterized by X-ray diffraction,transimission electron microscope,scanning electron microscope,X-ray photoelectron spectroscopy,Brunauer-Emmett-Teller adsorption isotherm.A possible forming mechanism of FeMoO4 hollow microspheres was proposed that lamellar FeMoO4 was generated and assembled into hollow microspheres on gas templates.FeMoO4 hollow microspheres had good adsorbing-reducing ability for Cr (Ⅵ) under visible light,with the reduction behavior of Cr (Ⅵ) following a first-order kinetics with the rate constant 2.15×10-2 min-1at pH of 2.The results of X-ray diffraction,inductively coupled plasma mass spectrometry and the cycling tests showed that the material was stable and consistently photoactive during use.

      Abstract:Single nucleotide polymorphisms (SNPs) of the porcine imprinting gene MKRN3 were identified in Meishan,Large white and Landrace pigs by PCR-RFLP and were genotyped in the F2 population of 285 Large white × Meishan pigs by PCR-BstUI-RFLP.Subsequently,correlation between the porcine gene MKRN3 polymorphism and porcine carcass traits were analyzed.The results showed that,the porcine MKRN3 gene had seven SNPs in the coding region,including 293C>G,361G>A,497G>A,630G>A,678T>C,1376C>T,and 1407T>A.Of which,the 678T>C site had different allele frequencies among domestic and foreigner pig species.In addition,the 678T>C site of the porcine MKRN3 was associated with internal fat rate and buttock fat thickness significantly (P<0.05).The 678T>C site had a dominant effect on internal fat rate (P<0.05),showing an increased internal fat rate with heterozygote form and had an additive effect on buttock fat thickness (P<0.05),showing a decreased buttock fat thickness with the additive of the 678C allele.Therefore,the 678T>C site of the porcine MKRN3 might provide a guiding value for the molecular breeding practice.

    • Isolation and identification of goat SKIP gene and its association with growth traits

      Abstract:In order to explore the effect of the goat SKIP gene on growth and development,and find the molecular markers associated with growth trait in goats,primers were designed according to the homologous sequence of the bovine SKIP gene.A 1 350 bp expressed sequence of the goat SKIP gene was amplified by RT-PCR,including the complete coding sequence,encoding 449 amino acids.Through sequence comparison and phylogenetic analysis,it was found that the SKIP sequences between goat and sheep had the highest similarity (99.52%).Physical and chemical characteristics of the SKIP protein were predicted by bioinformatic methods.The predicted SKIP protein has a calculated molecular weight of 52.0 ku and a pI of 5.18,and contains a typical phosphatidylinositol 5-phosphate phosphatase (COG5411) domain which is conserved among species.Most of the SKIP protein was located in the cytoplasm (60.9%) and nucleus (26.1%),which was verified by indirect immunofluorescence assay.However,there was a dynamic transfer of SKIP from the nucleus to cytoplasm during myoblast differentiation.A single-nucleotide polymorphism (SNP) G/A was detected in the second intron of the SKIP in the Boer and Macheng Black goat breeds.PCR-RFLP was used to genotype this SNP in 318 F2 hybrids of Boer ?? Macheng Black goats.The SNP was significantly associated with four traits including birth weight,chest circumference,cannon circumference and body height.Allele G seemed to be associated with fast-growth trait.

    • Embryonic and post-embryonic development of Triplophysa orientalis (Herzenstein)

      Abstract:The embryonic and post-embryonic development of Triplophysa orientalis (Herzenstein) were observed and described in detail under a microscope with a digital camera.The results showed that the mature unfertilized eggs of T.orientalis were viscous, ovoid, with a diameter of 0.97-1.11 mm, and characterized by tiny adhesive when encountering water.At a water temperature of (16±0.5)℃, the embryonic development lasted 132 h and 9 min, with a cumulative temperature of 2 113.6℃·h.Additionally, the embryonic development can be divided into seven physiological periods including blastoderm formation stage, cleavage stage, blastula stage, gastrula stage, neural stage and organ formation stage.The newly hatched larvae was (4.86±0.09) mm in length and the post-embryonic development lasted 34 days at a water temperature of (16±0.5)℃, which can be divided into the yolk sac and post larvae stage, lasting 9 and 25 days, respectively.

      Abstract:Fractions with high inhibitory activities on α-amylase and lipase were isolated from water extracts of Qingzhuan tea via ultra-filtration technology.Results showed that the trapped fraction of 5 ku ultra-filtration isolated from water extracts had strong α-amylase inhibitory activity.The trapped fraction of 100 ku ultra-filtration isolated from water extracts had strong lipase inhibitory activity.The α-amylase and lipase inhibitory activity of trapped fraction of 5 ku ultra-filtration isolated from the ethyl acetate fraction was higher than that of the ethyl acetate fraction.It is indicated that ultra-filtration combining with ethyl acetate extraction can more effectively isolate fractions with high α-amylase and lipase inhibitory activities from water extracts of Qingzhuan tea.

    • Effects of food matrix on content and antioxidant activity of tea polyphenols during simulated gastrointestinal digestion

      Abstract:Simulated gastric intestinal digestion model was used to evaluate effects of food matrixes on the contents and antioxidant activities of tea polyphenols in vitro.Results showed that adding milk,gelatinized starch and soybean oil significantly decreased content of tea polyphenols by 21.7%,15.0% and 3.3%,respectively.Comparing tea polyphenols after simulated gastric digestion,adding sucrose or citric acid had no influence on its content.DPPH·radical scavenging capacity of tea polyphenols significantly increased and ABTS+·radical scavenging capacity significantly decreased after adding citric acid,with no changes of the total antioxidant capacity.ABTS+·radical scavenging capacity of tea polyphenols significantly decreased after adding sucrose.Other antioxidant activities had no changes.The antioxidant activities of tea polyphenols were significantly reduced after adding gelatinized starch or milk.Soybean oil had no influences on its antioxidant activities.Comparing with tea polyphenols after simulated gastrointestinal digestion,adding milk significantly decreased contents of tea polyphenols by 7.7% and adding others had no influence.The total antioxidant capacity and ABTS+·radical scavenging capacity of tea polyphenols significantly increased after adding citric acid.DPPH·radical scavenging capacity had no change.DPPH·and ABTS+·radical scavenging capacity of tea polyphenols significantly increased after adding soybean oil,with total antioxidant capacity of no change.ABTS+·radical scavenging capacity of tea polyphenols significantly increased after adding gelatinized starch or milk,with other antioxidant activities of no changes.The antioxidant activities of tea polyphenols had no changes after adding sucrose.

      Abstract:The three-dimensional temperature and gas flow fields of transitional ventilation of closed henhouse with semi ladder coop were simulated with CFD software.In the simulation,the outdoor air temperature was 25℃ during the day and 13℃ during the night.The guide plates of side windows were provided with four different angles.Results showed that the angle of guide plate affected the uniformity of gas flow field and temperature field.The optimal angle of guide plates was found.The results of comparing the measured data with simulation showed that the relative error of gas velocity was 14.7% during the day and 11.5% during the night.The relative error of the temperature was 3.1% during the day and 3.6% during the night.The numerical simulation is in good agreement with the field measurement.It will provide a theoretical basis and guidance for the environmental control of the closed henhouse with semi ladder coop.

    • Rapid detection of calorific value in straw biochar based on FT-MIR

      Abstract:Five kinds of straw including cotton straw,wheat straw,corn straw,rape straw,rice straw collected by carbonization processing were used as straw biochar samples.Calorific value were measured with the C2000 heat meter and infrared spectrum were collected with the VERTEX type 70 Fourier infrared spectrometer.Leverage and residual was used to eliminate abnormal samples.SPXY was used to divide the sample set.TQ Analyst 8.3 was used to preprocess the spectra.The PLS model was established to predict the gross and net calorific value of straw biochar.The results showed that the multiple scatter correction (MSC) in combination with first derivative was the best model of preprocessing gross calorific value,with correlation coefficient of 0.909,verify the root mean square error of 614 J/g,and relative standard deviation of 2.61%.Multiple scatter correction (MSC) and first derivative in combination with the S-G smoothing was the best model of preprocessing net calorific value,with correlation coefficient of 0.873,verify the root mean square error of 707 J/g,and relative standard deviation of 3.12%.It is indicated that using FT-MIR to predict calorific value of straw biochar is feasible.
