Abstract:Ratoon stunting disease (RSD) of sugarcane (Sacchrum spp. Hyb.), caused by bacterium Leifsonia xyli subsp. xyli (Lxx), is one of the most important economically damaging diseases worldwide. The objective of the present study was to determine the effects of RSD on plant growth and the endogenous hormone levels in sugarcane stalks. Two mechanical cane varieties (i.e. sugar cane variety of ROC22 and chewing cane variety of Badila) were selected as the plant materials and RSD infection was determined by using PCR reaction. The seedcanes were grown in pots, and three treatments were done: pathogen-free seedcanes as control; seedcanes with thermal treatment: RSD infected seedcanes were treated at 52 ℃ for 30 min; RSD infected seedcanes. During the experiment, the plant heights were investigated, and the endogenous hormone levels in cane stalks were analyzed after planting in 90,120,150 and 180 d, respectively. The results showed that: the plant heights of the RSD infected seedcanes were significantly lower than those of the control and the thermal treatment. The contents of GA3 and IAA were obviously lower in the RSD infected seedcanes than those in the control and the thermal treatment in contrast to the content of ABA which was higher in the infected seedcane treatment as compared with the control and the thermal treatment; two cane varieties showed the same patterns and there was no significant difference between the control and the thermal treatment in plant height and endogenous hormone levels. Therefore, the thermal therapy is efficient to control RSD in sugarcane.

Abstract:To detect doxycycline (DC) residues in aquatic products,a ciELISA method were developed through preparing monoclonal antibody (McAb) by immunizing BALB/C mice, using doxycyline-bovine serum albumin conjugate (DC-BSA) as immunogen. The BALB/C mice used in the experiment for cell fusion were selected by two methods, the indirect ELISA and the indirect competitive ELISA.A hybridoma cell strain (3D1-H4),which can secret a McAb that specifically reacted with DC,were obtained. The McAb was separated by using hybridoma technology, and it was identified to belong to IgG1 by the indirect ELISA.A ciELISA for detecting DC was then developed, using this McAb.This McAb was characterized on its sensitivity,IC50 and veracity. The results showed that the titers of the McAb were 1∶(8×102) and 1∶(5.12×105) in supernatant and ascites, respectively. This McAb had 8.87%,5.86% and 2.74% cross-reactivity with tetracycline, oxytetracycline and chlotetracycline, respectively. The linear detection range for this ciELISA varied from 1.58 to 199.53 ng/mL,with a sensitivity of 2.88 ng/mL and an IC50 of 36.31 ng/mL.The recoveries of DC spiked in fish muscle and liver were 85.13% and 79.69%,respectively.Both the inter-assay and intra-assay coefficient variations were less than 15%.These results suggested that this ciELISA method was able to be used for detecting DC residues in aquatic products.

Abstract:In order to understand the water quality condition of Dacha Lake in Wuhan, composition of phytoplankton in the lake was investigated from June 2010 to May 2011.A total of seven phyla,59 genera and 89 species of phytoplankton were identified from qualitative and quantitative samples.Based on the species and quantity of phytoplankton, biological assessment of water pollution and eutrophication condition was conducted by using the methods of indicator organism and biotic diversity index (Margalef and Shannon-Weiner index). In the phytoplankton community,Chlorophyta and Cyanophyta were dominant in both alga species and quantities, followed by Bacillariophyta.Of the pollution-indicating phytoplankton, the algae indicating α-ms and β-α-ms types of eutrophication were dominant, accounting for 48.72% and 23.08% of the total, respectively. The Margalef index and Shannon-Weiner index for phytoplankton ranged from 1.45 to 2.19 and from 1.64 to 2.95,respectively.The results indicated that the water quality of Dacha Lake was of moderate pollution (α- or β- moderate pollution),and the trophic status was in the state of eutrophication (α-ms or β-α-ms),mainly in the type of α-ms as revealed by the pollution-indicating algae.

Abstract:The free amino acids and nucleotides were analyzedinraw and cooked silver carp (Hypophthalmichthys molitrix) musclesof different body weight (1.0-3.0 kg) using TAV (taste active value) and EUC (equivalent umami concentration) methods. The results showed that Tau,His and Gly were the main free amino acids in silver carp muscle, and their concentrations decreased after cooking.Histidine significantly contributed to the bitter taste,Glu contributed to the major umami taste, while Gly and Ala contributed to the sweet taste. The nucleotide concentrations in fish muscles of different body weights mainly dominated by IMP and HxR,and there were significant differences in nucleotide concentrations between raw and cooked muscles. The TAV of IMP and AMP decreased after cooking, while the TAV of Hx increased, and the major umami taste was still contributed by IMP.The EUC showed a trend of increasing followed by decreasing, with increasing body weight. And it reached the maximum (31.57 g MSG/100 g) at 2.0 kg for cooked fish muscle.

Abstract:A remote control traction monorail transporter 7YGDQ-50 was designed specifically for the complex mountainous environment. The transporter is composed of the electric hoist, the telecontrol, the control box, the driving wheels, the wire rope, the guide wheel, the travel switch, the trailer car, the track and the adjustable counterweight device, etc. To solve the problems of the present traction monorail transporter, the key components, which include the electric hoist, the control box, the driving wheels, the trailer car and the adjustable counterweight device, were redesigned and improved by the change of the driving mode. The transporter can move uphill, downhill or park by the telecontrol or a manual button. The field experiments show that the transporter improves the work efficiency compared with the existing traction monorail transporter, and is suitable for the complex mountainous transport operations. While the telecontrol is used, the receiving device should be in visual range.

Abstract:The activity sounds of Sitophilus zeamais adults in wheat, corn and Sitophilus oryzae larvae in coix seed were respectively sampled in different temperatures, and decomposed by wavelet packet. Three characteristics of decomposition nodes were analyzed. They were nodes energy, nodes energy ratio and energy entropy. The research found that the nodes energy and nodes energy ratio of the activity sound of Sitophilus zeamais adults in wheat and corn were greatly different in the same temperature, and the difference of frequency distribution between these two types of signals was obtained. It was also found that there was much difference between the energy entropy of these two types of signals, and the activity acoustic signal energy entropy mean statistical data of Sitophilus zeamais in wheat and corn were collected. In different temperatures, the nodes energy, nodes energy ratio and energy entropy of the activity sound of Sitophilus oryzae larvae in coix seed were also different. The results show that both the difference of temperature and grain species affect the activity sound characteristics of grain insects, and the activity sounds in wavelet packet decomposed characteristics are significant different.
