Abstract:为探讨血管紧张素Ⅱ(angiotensin Ⅱ,AngⅡ)对3T3-L1前脂肪细胞分化的影响,采用鸡尾酒法诱导3T3-L1前脂肪细胞分化,并用油红O染色法和还原型烟酰胺嘌呤二核苷酸(nicotinamide-adenine dinuceotid,NADH)氧化速率法检测了3T3-L1脂肪细胞分化过程中胞浆脂质的累积和甘油磷酸脱氢酶(glycerol phosphatedehydrogenase,GPDH)的活性.结果表明,100 nmol/L的AngⅡ可显著增大3T3-L1前脂肪细胞分化过程中胞浆脂质累积和GPDH活性(P<0.01),外源性AngⅡ可促进3T3-L1前脂肪细胞的分化,增加脂肪细胞中脂类合成和储存.

Abstract:将35个不同基因型的马铃薯试管苗单节段接种在添加20 μmol/L嘧啶醇的MS培养基中,在光照16 h/d和(17±1)℃温度条件下培养6个月后,分析嘧啶醇对试管苗生长的影响.结果显示,除基因型CE76外,20 μmol/L嘧啶醇对于参试基因型试管苗生长速度均有显著的延缓作用,但在基因型间存在较大差异.80%左右的基因型在添加嘧啶醇的培养基中培养6个月后其试管苗株高低于8 cm,其抑制率在50%以上.添加嘧啶醇培养基中培养的试管苗长势健壮,节间距显著缩短而节数没有显著减少,嘧啶醇也强烈地抑制根伸长.

Abstract:采用胡萝卜片愈伤组织接种培养法,在室内条件下对供试香蕉穿孔线虫10个种群的耐寒性进行了测定.结果表明,在12℃和10℃低温下分别处理15 d、25 d、35 d后,供试香蕉穿孔线虫各种群生活力虽然有所下降,但仍然能够保持一定的繁殖能力;而在8℃时,各种群生存能力明显下降,有些种群无法在此温度下存活.8℃处理15 d后,Rs6种群无法存活;处理25 d后,Rs 2种群亦无法存活;处理35 d后,Rs8、Rs4种群无法存活.Rs1、Rs3、Rs5、Rs7、Rs9和Rs10等6个种群在8℃低温下处理35 d后仍然能够存活.

Abstract:采用药膜法、滤纸药膜法和饲料拌药法分别检测β-细辛醚对玉米象成虫的触杀、驱避及种群抑制等作用.结果表明,β-细辛醚对玉米象成虫具有良好的防治效果.β-细辛醚157.30、78.63 μg/cm2浓度处理玉米象成虫96 h后的校正死亡率分别高达100.00%和93.22%,处理96 h后的触杀毒力LC50为28.22 μg/cm2;157.30、78.63、39.32 μg/cm2浓度处理的β-细辛醚对玉米象成虫的平均驱避率在V级以上,较低浓度19.66 μg/cm2处理的驱避率也达到了Ⅳ级;0.500 g/kg的浓度处理7 d后,玉米象成虫的校正死亡率达到91.11%,处理45 d后,对子代种群抑制率也高达100.00%.

Abstract:利用丁香假单胞菌(Pseudomonas syringae)冰核基因的N-末端(inpn)作为锚定单元(anchoring mo-tif),通过与来自黄杆菌属(Flavobacterium sp.)的有机磷水解酶基因(opd 构建融合基因inpn-opd,并连接于假单胞菌表达载体Pymbp,然后导入恶臭假单胞菌(P.putida)野生型菌株AB92019,获得了能在其细胞表面展示有机磷水解酶并具有全细胞酶催化活性的重组工程菌MMBL-opd.SDS-PAGE结果表明,融合基因能表达产生80 ku的蛋白质.重组菌MMBL-opd在无抗性LB固体培养基上能稳定生长,所携带的外源质粒的稳定性达到100%;在添加100 μmol/L Go2 培养基上28℃培养48 h,表面展示的有机磷水解酶具有最高全细胞酶活性,为0.036 U/mg.用蛋白酶K消化处理重组菌表面蛋白可使其全细胞酶活降低92%.重组菌在PBS缓冲液中于4℃条件下保存30 d,仍能保持93%的全细胞酶活.

Abstract:构建了鸭MHCⅡα的融合表达载体PET-28a,对融合蛋白在大肠杆菌中表达量的影响因素进行了研究.SDS-PAGE分析表明,诱导温度22℃,时间3 h,IPTG浓度0.05 mmol/L,OD600m值0.45,通气量90%和pH 7.2时MHCⅡα融合蛋白的表达量最高,占菌体总蛋白的23.4%,比优化前提高了16.7%.蛋白经纯化后纯度可达90%.Western blot分析表明,该蛋白可被鼠抗鸭MHCⅡα阳性血清识别,具有较好的免疫学活性.

Abstract:对草鱼受精卵及所处水体的好氧及兼性厌氧菌的数量和组成进行了研究.结果表明:草鱼受精卵菌群的数量随受精卵的发育呈递增趋势,刚受精时每个受精卵的细菌数量为1.24×101cfu,出膜前增加到1.41×105cfu.水体中细菌的数量相对稳定,为1.05×104~3.85×104cfu/mL,只在受精卵出膜前的阶段急剧增加,为1.32×106cfu/mL.随受精卵的发育,受精卵的优势菌群先为气单胞菌属(Aeromonas)、肠杆菌科(Enterobac-teriaceae)、微球菌属(Micrococcus)和棒杆菌属(Corynebacterium),后为莫拉氏菌属(Moraxella)和不动杆菌属(Acinetobacter).水体中的优势菌群为气单胞菌属(Aeromonas)、肠杆菌科(Enterobacteriaceae)和莫拉氏菌属(Moraxella).受精卵的优势菌群与其所处水体的优势菌群基本相同.

Abstract:以盾叶薯蓣的叶片制备ISSR-PCR DNA 模板.通过单因子试验分别研究了模板DNA质量和浓度、Mg2 浓度、Taq酶用量、dNTPs浓度以及退火温度对盾叶薯蓣ISSR-PCR扩增的影响,建立了适宜于盾叶薯蓣的ISSR反应体系和扩增参数,即15μL反应体系中包括:20 ng/L模板DNA,1×Taq酶缓冲液,1.0 U Taq聚合酶,2.5 mmol/L MgCl2,0.2 μmol/L引物和0.3 mmol/L dNTPs.并从46个ISSR引物中筛选出10个带纹清晰、多态性丰富的引物,并确定了这些引物的适宜退火温度.

Abstract:以水曲柳和落叶松为研究对象,在一个生长季内采用随机土芯法研究了林地土壤速效氮和细根的空间分布变异以及2树种细根对速效氮的觅食行为.结果表明:水曲柳和落叶松细根各参数(生物量,根长密度和比根长)及土壤速效氮(铵态氮和硝态氮)在林地土壤中呈异质性分布,水曲柳细根各参数平均值显著高于落叶松,但2树种细根的变异系数一致.2树种细根生物量和根长密度均与2种形态氮呈显著正相关,但水曲柳细根与2种形态氮相关性高于落叶松,而且硝态氮对2树种细根生物量和根长密度分布的影响大于铵态氮,比根长与速效氮没有显著相关性.以上研究结果表明,2树种具有较强通过在富养的土壤斑块增生而有效觅食的能力,但水曲柳细根的这种增生能力大于落叶松;落叶松细根除具备一定形态可塑性外,可能还通过提高其吸收速率以及菌根数量有效觅食.

Abstract:卡罗来纳杜鹃是一种优美的常绿灌木,原产美国东部沿海并能耐-33℃的低温.由于该植物无性繁殖技术困难,限制了其及其栽培变种在园林中的应用.于10月份采集卡罗来纳杜鹃的木质化枝条,在美国缅因大学的植物繁殖温室中进行扦插繁殖.处理后的插穗被随机放在具有恒温(24℃)低温加热的苗床上.结果表明:IBA对卡罗来纳杜鹃木质化插条的生根有显著影响,IBA处理后的插条生根率为25.0%~84.4%,对照没有生根.IBA浓度对插条生根也有显著影响,处理Ⅳ即先快蘸液态KIBA(8 000 mg/L)再迅速蘸取粉状IBA(8 000 mg/L)的插条生根率最高为84.4%;当IBA浓度≤8 000 mg/L时,生根率≤43.8%;液态.KIBA 16 000mg/L 的浓度太高会使插穗出现烧死现象;当KIBA浓度为200 mg/L和400 mg/L时,浸泡插穗24 h后生根率分别为31.3%和37.5%.IBA的类型及处理浓度对根质量(根体积)有显著影响,混合处理Ⅳ的生根质量最好,体积为38.9cm3;KIBA浓度200 mg/L时浸泡24 h后处理的插穗生根体积最小,为12.1 cm3.生根插穗移人低温储藏8周后,可移栽和培育出圃.

Abstract:针对鲜切莲藕褐变问题,以刀片刃角为主要试验因素,探讨通过刀片刃角与莲藕切割阻力的关系,并以刀片刃角、刀片滑切角、刀盘转速为主要试验因素,研究回转式切割器主要性能参数对莲藕褐变度的影响.结果表明:刀片刃角由10°增大到20°时,切割力的增加幅度较小,而当刀片刃角从20°变为25°时,切割力曲线几乎呈直线上升;建立了刀片刃角与切割力关系回归模型,经验证计算值与实测值拟合良好;3个因素对褐变度影响的主次顺序为刀片刃角(γ)>刀盘转速(n)>滑切角(τ),且刀片刃角越小,刀盘转速越大,滑切角越大,莲藕褐变度越小.
