Abstract:从广东省韶关和梅州2个烟草产区采集了来自8个县市的共38个烟草青枯菌菌株,对病菌的生物型、致病型和遗传多样性进行了测定,结果表明广东烟草青枯菌全部为生物型Ⅲ。通过在红花大金元,K326,岩烟97和系3等4个烟草鉴别品种上的致病性反应,将广东烟草青枯菌分为强致病力、中等致病力和弱致病力3种致病型,其中以中等致病力菌株为主,占78.9%;强致病力菌株占13.2%;弱致病力菌株占7.9%。从116个RAPD通用引物中筛选出10条引物,用于对38个烟草青枯菌株DNA的RAPD分析,扩增条带表现出明显的多态性,条带主要聚集在0.3~4.0 kb之间。聚类分析这38个菌株可分为2个组群,即组群A和组群B。组群A中又可以分为7个亚组(A1、A2、A3、A4、A5、A6、A7);组群B也含有4个亚组(B1、B2、B3、B4)。研究结果还表明,广东烟草青枯菌菌株RAPD组群的划分与菌株的寄主来源有一定的相关性,但与地理区域、生物型和致病型没有明显的相关性;生物型不能反映烟草青枯菌菌系的差异。

Abstract:研究了培养温度、培养时间、培养基pH值以及异丙基硫代-β-D-半乳糖苷(isopropylthio-β-D-galacto-side,IPTG)浓度等不同条件对鸡毒霉形体黏附素截短蛋白在大肠杆菌中表达量的影响。经SDS-PAGE分析表明:诱导温度为28℃、诱导时间为9 h、培养基pH值为7.0、IPTG浓度为0.010 mmol/L时目的蛋白在细菌裂解沉淀和上清液中均最大量表达,分别达到30%和17%左右。上清液经GST.Bind Resin亲和层析分离纯化后,洗脱产物中pMGAⅣ融合蛋白纯度高达95%。Western-blot分析表明,该蛋白可被鸡毒霉形体阳性血清识别,具有较好的免疫学活性。

Abstract:将编码弓形虫微线蛋白3(MIC3)的真核表达质粒pcMIC3和编码γ-干扰素(IFN-γ)的真核表达质粒pcIFN-γ各100μg混合肌肉注射,间隔2周、3次免疫Balb/c小鼠,同时设相同剂量的pcMIC3、pcDNA和PBS免疫对照组。分别以酶联免疫吸附试验(ELISA)、四氮甲唑蓝(MTT)比色法和乳酸脱氢酶释放法(LDH)检测试验小鼠血清的特异性抗体、脾淋巴细胞的增殖应答和细胞毒性T细胞(CTL)活性,并观察腹腔接种弓形虫RH株速殖子后质粒的免疫保护效果。结果表明:与其它试验组相比,pcMIC3 pcIFN-γ组试验小鼠在免疫8周后的ELISA抗体水平、脾淋巴细胞的增殖应答和CTL活性显著或极显著提高,免疫小鼠45 d时的生存率为100%,能有效抵抗小剂量弓形虫强毒株的攻击。

Abstract:在纯化日粮中分别添加0、60、120、180、240和300 mg/kg还原型谷胱甘肽(GSH),饲喂平均体重为(1.12±0.01)g的凡纳滨对虾(Litopenaeus vannamei)8周,观察饲料中不同浓度谷胱甘肽对凡纳滨对虾非特异性免疫因子和部分相关酶活性的影响。结果表明:随着饲料中GSH添加量的增加,凡纳滨对虾的髓过氧化物酶(MPO)、溶菌酶(LSZ)、碱性磷酸酶(AKP)、酸性磷酸酶(ACP)活性值均呈先升后降的趋势,在120~240 mg/kg时达到最高值,且显著高于对照组;随着饲料中GSH添加量的增加,血清和肝胰腺中的谷草转氨酶(AST)和谷丙转氨酶(ALT)呈下降趋势,在240 mg/kg组达到最低,且显著低于对照组,但在300 mg/kg组又有所回升。以上研究表明,饲料中添加一定浓度的GSH能有效提高凡纳滨对虾血清和肝胰腺中髓过氧化物酶、溶菌酶和磷酸酶活性,并影响转氨酶的活性,从而激发了凡纳滨对虾的非特异免疫功能。

Abstract:对分离到的1株异养型绿球藻(Chlorococcum sp.)的碳源、氮源及其它培养特性进行了研究,并分析了该藻种在不同氮源及其水平下的氨基酸组成和含量。试验结果表明:该绿球藻异养培养的最佳碳源和氮源分别为葡萄糖和尿素;最适pH值为7.0~8.0;最适培养温度30℃左右,最适异养培养条件下的倍增时间为5 h左右。在含1倍和2倍氮浓度的培养条件下,绿球藻的粗蛋白含量分别达30.8%和36.6%。相同氮浓度下,氮源对氨基酸组成和含量无明显影响。

Abstract:定期测定了储良龙眼(Longan chinensis Sonn.cv.chuliang)果实在不同贮藏条件下的纤维素酶活性,并探索其与果肉自溶的关系。结果表明:常温和丙烯处理后,随着贮藏时间的延长,果肉纤维素酶的活性逐渐增强,最大值分别出现在第4天和第5天,果肉自溶指数也随之增高,二者的变化趋势一致。4℃下贮藏1个月时,纤维素酶活性变化较小,始终保持在100μg/(mL.h)左右,此条件下贮藏的果实只在贮藏后期开始自溶,但在之后的货架期纤维素酶活性迅速升高,24 h就达150μg/(mL.h),自溶指数也迅速提高。

Abstract:对柳叶桂、山桂、四季桂3个品种枝条的生长、花量、叶片水分状况、叶绿素含量和光合作用等的年周期变化进行了测定。结果表明:3个品种的总生长量和鲜花产量差异显著,总生长量为柳叶桂>山桂>四季桂,而全年鲜花产量为柳叶桂>四季桂>山桂。在年周期变化中,叶片组织水和自由水含量不断减少,束缚水含量不断升高。叶绿素含量呈先上升后下降的趋势,8月份的含量最高。4、6、8、10四个月光合作用的日变化呈双峰曲线,其中4月的峰值出现在12:00和16:00左右,其它3个月出现在10:00和16:00左右,第1峰值均比第2峰值高;12月份为单峰曲线,峰值出现在14:00左右。Pn随光合有效辐射增加而增加,但光合有效辐射增加到1 600~1 800μmol/(m2.s)时,Pn增加不显著。随着温度的升高,Pn呈先上升后下降的趋势,在28℃时Pn达到最大值。最大光化学效率日变化呈单峰曲线,但呈现先下降后上升趋势。

Abstract:采用常温贮藏、泡沫箱加冰贮藏和低温贮藏3种方式,观察其对采后新鲜生菜食用品质的影响。结果表明:生菜采收后,感官品质随贮藏时间的延长而下降,失重率和亚硝酸盐含量上升;低温贮藏能明显减缓生菜的失重率和亚硝酸盐的生成量,降低呼吸强度,推迟呼吸高峰的出现;泡沫箱加冰贮藏的效果差于低温贮藏,略好于常温贮藏。

Abstract:生物柴油可以由菜籽油与甲醇在碱催化剂的作用下通过酯交换反应制得。为解决生物柴油酯交换过程中的产物与催化剂分离问题,制备了负载型固体碱催化剂,观察了反应条件如醇油比、催化剂用量、反应温度、反应时间等的变化对生物柴油转化率的影响。采用正交试验方法找出菜籽油酯交换反应的最佳反应条件为醇油摩尔比18∶1,催化剂用量8%,反应温度65℃,反应时间7 h。在此反应条件下生物柴油转化率可达到98%以上。
