Abstract:以水稻红莲型细胞质雄性不育(HL-CMS)不育系粤泰A(YTA)和保持系粤泰B(YTB)黄化苗线粒体为材料,研究了YTA和YTB离体线粒体KA TP通道对其诱导调节剂KCl、ATP、ADP和GTP等的响应特性。结果表明,KA TP通道诱导剂KCl,能明显诱导YTA和YTB线粒体膨胀,但YTA的膨胀程度较YTB的明显;KA TP通道的内源性抑制剂ATP对YTA和YTB线粒体膨胀起显著抑制作用;KA TP通道的内源性激动剂ADP和GTP引起的不育系YTA离体线粒体短暂收缩及之后的再膨胀程度均较YTB的明显;此外,比较YTA和YTB离体线粒体在Rh123一起孵育10 min后,KCl和解偶联剂FCCP处理引起的离体线粒体膜电位(Δψm)下降过程,发现前者的线粒体Δψm较后者的易于过早崩解。对水稻HL-CMS不育系YTA和可育的保持系YTB线粒体KA TP通道特性的比较研究表明,水稻HL-CMS不育系YTA线粒体KA TP通道对其诱导调节剂KCl、ATP、ADP和GTP等的响应较YTB的更敏感。

Abstract:在江苏沿江地区,以迟熟中粳品种通育粳1号为材料,研究了270 kg/hm2施氮量、有机氮肥占比40%作基肥施用情况下,分蘖肥和穗肥氮素配比对水稻产量、稻米品质及其氮肥利用率的影响。结果表明:生育前期氮肥(蘖肥)比例提高,单位面积穗数增加,每穗粒数减少。在早施分蘖肥情况下,产量以蘖肥占比20%>10%>30%;蘖肥和穗肥不同配比对糙米率、精米率、整精米率、垩白率等品质指标的影响差异不显著,不同处理间的稻米直链淀粉含量存在较大差异,稻米蛋白质含量随穗肥比例提高而增加,当穗肥占50%时,稻米淀粉RVA谱出现显著变化,主要表现为峰值粘度降低、崩解值减少、消解值增大和糊化温度提高;大田促蘖和保蘖肥各占10%、促花占30%、保花占10%时,有利于协调水稻产量和稻米品质,同时具有较高的氮素利用率。

Abstract:选用湖北省主栽水稻品种88份,分别在崇阳、远安、武汉三地进行田间自然诱发稻瘟病和纹枯病。抗性鉴定试验结果表明,武汉地区未发生稻瘟病,崇阳、远安两地88个品种除武运粳7号外几乎全部发病,发病率达98.86%。抗稻瘟病的早、中晚稻品种数分别为14份和23份,分别占参试品种的45.16%和40.35%。崇阳、远安、武汉三地综合分析显示,88份水稻品种中抗纹枯病品种13份,占参试品种的14.77%。

Abstract:以多杀菌素产生菌C121菌株为试验菌株进行发酵培养基的研究,探索不同碳源、氮源对多杀菌素生物合成的影响。经L18(37)正交筛选试验,最终筛选到一个发酵培养基的最佳配方。用菌株C121进行摇瓶发酵条件试验的结果表明:当500 mL三角瓶中装料40 mL,消前pH 7.0,转速为210 r/min,温度为30℃时是最佳发酵条件,其发酵效价达到586μg/mL。

Abstract:以高产S-腺苷甲硫氨酸的菌株Saccharomyces cerevisiaeB9对培养基和发酵条件进行了优化研究。通过单因素水平测试和正交水平测试,确定了发酵培养基的最佳配方组合为:蔗糖8%、NH4Cl 0.5%、NH4H2PO40.2%、酵母提取物1.2%、MgSO4.7H2O 0.01%、KH2PO40.6%、L-甲硫氨酸1.2%,与原始发酵培养基相比,S-腺苷甲硫氨酸从1.16 g/L提高到3.35 g/L,菌体干重从16.18 g/L提高到18.25 g/L。在此基础上,对发酵条件进行优化,结果发现:初始pH5,培养温度30℃,摇床转速180 r/min为最佳培养条件。此时,菌体干重达19.86 g/L,S-腺苷甲硫氨酸产率达4.2 g/L。

Abstract:通过RT-PCR获得猪Ghlelin基因cDNA片段,克隆入pcDNA3.1( )质粒中构建真核表达质粒pcDNA-pGhl,并将该质粒注射到大鼠和新西兰兔后腿肌肉中,以研究pGhlelin真核表达质粒对动物生长和血液相关激素分泌的影响。大鼠试验表明:pcDNA-pGhl在注射初期的1个月内有增加大鼠体重及在整个试验期有降低料重比的趋势。在注射质粒后第6天,试验组个体增重提高10.6%,差异显著;饲料转化率提高10%,差异极显著。新西兰兔试验表明:试验组血液GH水平一直高于对照组,且在注射后2周高出35%,同时随着GH浓度的增加,血液中SS的浓度相应升高。注射重组质粒pcDNA-sGhl有增加血浆GH和SS水平趋势。以上结果表明,Ghrelin基因真核表达质粒通过促进GH的分泌促进生长,但其促生长效能较短暂,可能是由于同时促进生长抑素分泌。

Abstract:有报道,兔肉内矿物质总量是1.779%,其中磷的含量为0.579%;成年动物体中含磷量按新鲜组织计算为0.60%~0.75%,按干组织计算为1.9%~2.5%。传统的家兔体内磷含量测量方法是通过宰杀后取样,用原子吸收光谱法(AAS)和离子质谱仪(SIMS)测量以及用反应堆、加速器产生的中子源辐照样品进行活化法分析测量。这些方法都是离体测量的,且是动物器官取样。

Abstract:建立鸡蛋、牛奶中甲硝唑残留检测的高效液相色谱法(HPLC)。样品中甲硝唑用乙酸乙酯提取、浓缩后用正己烷-乙酸乙酯(2∶1)溶解,Silica(SiO2)固相萃取柱净化,氮气吹干,流动相溶解,HPLC测定。固定相为C18色谱柱,流动相为pH 4.3醋酸缓冲液∶乙腈(88∶12,V/V),紫外检测波长325 nm。该方法可检出鸡蛋、牛奶中甲硝唑的最低定量限为1μg/kg,在空白样品添加浓度为1μg/kg时,鸡蛋、牛奶中甲硝唑的平均回收率分别为69%和75%,日间变异系数均小于15%。产蛋鸡连续饲喂含甲硝唑(250 mg/kg)饲料1周,用所建方法可检测到停药后14 d内鸡蛋中原药及代谢物残留。结果表明,该方法的样品处理方法简单,最低定量限、回收率和变异系数符合兽药残留检测方法要求。

Abstract:研究紫杉醇及其重要前体物质巴卡亭Ⅲ在中国红豆杉(Taxus chinenesis)中合成和积累部位,通过HPLC分析代谢产物在中国红豆杉植物中的空间分布,定量PCR技术研究代表紫杉醇合成途径中不同步骤的6种关键酶基因在植株中的表达特征。结果表明,巴卡亭Ⅲ主要分布在针叶中,而紫杉醇主要分布在树皮和树根皮中;针叶中,紫杉醇代谢途径上游步骤的关键酶基因表达水平较高,树皮中催化紫杉醇侧链连接的酶转录表达水平高,而树皮中能检测到较高含量的6种关键酶基因的表达。表明紫杉醇的骨架结构主要在针叶中合成,而树皮中转化这些前体物质生成紫杉醇的能力较强,树根中含有完整的紫杉醇代谢酶系,暗示根组织培养是生产紫杉醇很有前景的方法。进一步建立了组织扦插培养红豆杉离体组织体外生产紫杉醇的系统。在茉莉酸甲酯的诱导下,红豆杉针叶在体外能够生产紫杉醇;树皮中由于缺乏足够的前体物质,离体培养中紫杉醇含量不断下降;枝叶培养中,产生的紫杉醇主要集中在树皮中。上述研究结果暗示,紫杉醇及其前体物质在中国红豆杉体内存在转移现象。

Abstract:比较了用乳酸、酸化亚氯酸钠(ASC)、次氯酸钠(NaClO)配制的保鲜液对冷却猪肉的保鲜效果。对猪肉进行浸泡处理后在5℃冷藏条件下存放7 d,分别于处理后0 d、3 d5、d、7 d对猪肉及对照样品进行微生物指标、TVB-N等方面的测定。结果表明,这几种保鲜液对猪肉都有一定的抑菌保鲜效果。

Abstract:利用棚面与地面自然高差,设计了温室棚面雨水集蓄及自压式渗灌系统,并通过对比试验探讨了雨水自压式渗灌和浇灌条件下温室蔬菜生长环境状况。试验结果表明:冬季集雨渗灌与浇灌相比,晴天全天平均相对湿度降低8.5%,阴雨天全天平均相对湿度降低6.5%;室内空气温度比浇灌平均高出1~3℃;表层土壤平均温度比浇灌低0.26℃,但20 cm深度土壤平均温度比浇灌高0.64℃;全生育其渗灌总灌水量为浇灌的40%,收获蔬菜(包青包)平均高度增加4.3 cm。
