Abstract:RAPD(random amplified polymorphic DNA)即随机扩增多态性DNA技术是由Williamsc和Welsh发展起来的一项研究遗传物质本身的分子生物学技术,克服了从表型、染色体和蛋白质水平研究物种遗传多样性所固有的缺点,并具有相对简便、易操作、省时省力、无需设计引物和产物遗传多样性丰富等优点,已被广泛地应用于生物研究中。笔选取了3个鸭种,用RAPD技术研究其基因组DNA的多态性,目的在于深入了解其种质、亲缘关系及遗传多样性,为保护鸭种质资源和分析鸭配套品系的遗传多样性提供分子生物学方面的证据。

Abstract:利用PCR技术从弓形虫RH株基因组中扩增出SAGⅠ部分基因片段,亚克隆入表达载体pGEX-KG,将重组质粒转化入大肠杆菌BL21,IPTG诱导表达,SDS-PAGE电泳显示表达的重组蛋白的分子量为56ku。免疫印记分析显示此表达产物能被猪抗弓形虫阳性血清所识别,表明该重组蛋白具有免疫反应活性。为进一步进行弓形虫病的免疫预防和诊断研究奠定了基础。

Abstract:从柑橘果实分离的假丝酵母Candidaspecis CWW-4菌株对贮藏柑橘绿霉病菌有抑制作用。试验结果表明,拮抗酵母菌生防效果与病原菌和绿霉菌的浓度以及接种时间有关,在25℃下当接种绿霉菌浓度为1×104个/mL时,1×108cfu/mL拮抗酵母能较好的抑制绿霉病的发生。假丝酵母先于病原菌24 h接种,5 d后可完全抑制绿霉病的发生。在4℃和25℃下,拮抗酵母菌接种1 d以后可在果实的伤口处快速繁殖,伤口处无论是否接种病原菌其数量都分别增加20倍和60倍以上。采用不同的处理液处理果实,发现假丝酵母悬浮液和培养原液对绿霉病菌有较好的抑制效果;培养滤液和热杀死液抑制效果差。25℃离体条件下,1×108cfu/mL假丝酵母悬浮液24 h能完全抑制绿霉菌孢子的萌发,培养滤液和热杀死液无抑制效果。NYDB1所含各组分的营养液都不同程度的降低假丝酵母对绿霉病菌的抑制效果。研究结果表明,假丝酵母Candidaspecis CWW-4菌株对柑橘绿霉病有很好的防治潜力,其抑制机制可能与营养竞争有关。

Abstract:在不同压力(22 MPa2、4 MPa2、6 MPa2、8 MPa)和不同温度(30℃、45℃、60℃)条件下,用CO2超临界流体对非洲山毛豆进行萃取。结果表明:提取物中鱼藤酮的含量最高是在30℃和24 MPa的条件下为7.41%;每克干粉中提取出来的鱼藤酮质量最高的是在60℃和22 MPa的条件下,为4.49 mg。加入夹带剂后,提取物中鱼藤酮含量和每克干粉中提取出来的鱼藤酮质量最高的均在60℃和22 MPa时,加入10%(V/M)乙酸乙酯作夹带剂,结果分别为7.96%和5.99 mg。

Abstract:采用乙腈提取,ENVI-18固相萃取,丙酮洗脱,C18高效液相色谱分离柱,甲醇-水(体积比85∶15)流动相,紫外检测波长275 nm,建立了同时测定蔬菜中甲基对硫磷和对硫磷残留量的固相萃取-高效液相色谱法。结果表明:测定甲基对硫磷和对硫磷农药残留量的线性范围为0.05~5.00 mg/L,相关系数0.999 8~0.999 9,检出限0.025~0.031 mg/L,加标回收率81.60%~93.60%,相对标准偏差5.3%~2.9%。

Abstract:考察吡喹酮在不同溶媒中的溶解度、稳定性和对小鼠的刺激性,研制出了200 mg/mL的吡喹酮注射剂。用此注射剂对人工感染日本血吸虫22 d后的小鼠进行了肌肉注射治疗试验。结果表明:200 mg/mL吡喹酮注射剂对日本血吸虫病小鼠的治疗效果良好,减虫率可达100%。该注射剂在室温自然条件下避光保存8个月或在80℃恒温24 h,其颜色及吡喹酮的浓度均无变化。小鼠注射给药部位没有炎症和组织坏死变化。

Abstract:以70%饱和硫酸铵盐析法制备鳗源创伤弧菌生物1型高致病菌株FJ03-X2的胞外产物(ECPs),攻毒试验表明,该ECPs对鳗鲡无致病力;SDS-PAGE分析表明,该ECPs由约36 ku为主的系列蛋白条带组成。制备ECPs的免疫刺激复合物(ISCOMs),以20μg/尾的剂量腹腔注射免疫规格为15~20 g/尾的欧洲鳗鲡,ELISA分析表明,21 d和28 d时的血清抗体效价约为1∶1 280,免疫印记显示,ECPs中分子量为36 ku以上的蛋白成份可被免疫欧洲鳗鲡血清所识别,是构成ECPs的主要免疫原;免疫保护试验表明,免疫欧洲鳗鲡显示出较高的免疫保护力。试验结果也显示出,单纯以ECPs等剂量腹腔注射免疫欧洲鳗鲡,不能激发欧洲鳗鲡产生较高滴度的血清抗体,免疫欧洲鳗鲡也未能产生有效的保护性免疫应答。

Abstract:2002年每季度自道观河水库采样1次,每次采细鳞鲴(Xenocypris microlepisBleeker)鲜鱼10尾,测量体长、体重,取鳞片鉴定年龄。测定背部肌肉水分、粗蛋白、粗脂肪、粗灰分、碳水化合物等的含量,按Brett andGrove法计算能值。结果表明细鳞鲴肌肉中水分含量为76.38%,干物质粗蛋白含量为17.80%,粗脂肪含量为3.54%,粗灰分含量为1.79%,碳水化合物含量为0.49%;能值为5.91 J/mg(湿重)。各项生化指标和能值无明显季节变化;含水量随鱼年龄的增长而下降,能值、蛋白质、脂肪含量均随年龄增长而增加;能值的增长趋势在3龄前明显,3龄后趋于平缓,可以定性地认为其能量增长的“拐点”年龄为3龄左右。粗蛋白、粗脂肪含量以及能值与鱼体体长、体重都有显著相关关系,且各对相关关系均以二次方程Y=a bx-cx2的相关性最好。能值Q(%)与水分(Wat)含量的回归方程为Q=70.854 3?1.422 0(Wat) 0.007 5(Wat)2(r=-0.965 9)。通过回归方程以水分含量来估计粗蛋白质、粗脂肪含量和能值是完全可行的。

Abstract:为确定壶状臂尾轮虫休眠卵的最适形成和最适孵化条件,对不同温度、pH值和光照条件下壶状臂尾轮虫的休眠卵进行了促产和孵化研究,结果表明,外界环境因子的刺激是诱导轮虫两性生殖现象发生的直接原因:pH为7.5时壶状臂尾轮虫的休眠卵产量最大,混交雌体百分率也最高;30℃时休眠卵孵化率最高;光照对壶状臂尾轮虫休眠卵的形成和孵化并无显著影响。
