Abstract:puroindoline a (Pin a)和puroindoline b(Pin b)是控制小麦籽粒硬度的主效基因。根据已报道的小麦Pin a基因的保守序列,设计合成了1对特异性引物ForA1和RevA1,对六倍体牡山羊草Aegilops juvenalis(UUMMDD)的基因组DNA和胚乳cDNA进行Pin a基因扩增、克隆、序列测定和表达分析,发现了1个新型Pin a(Pin a~allele),其ORF长4/17bp,编码148个氨基酸残基,具有麦类作物Pin a基因特有的28个氨基酸的信号肽序列和WRWWKWWK的色氨酸结构域基因序列,与软粒小麦cv.capilole的Pina—Dla相比较,其核苷酸同源性和氨基酸同源性分别为98.7%与96.6%。Pin a~allele含有1个紧邻色氨酸结构域的突变位点(Gln 77Leu)。RT~PCR证实了Pin a在籽粒胚乳中的表达。Soulhern Blot分析结果表明,牡山羊草中Pin a基因含有1个拷贝。研究结果表明,山羊草中包含着与小麦差异较大的籽粒硬度控制基因。

Abstract:按金黄色葡萄球菌多重耐药转运蛋白NorA的编码序列设计引物,以金黄色葡萄球菌基因组DNA为模板,扩增出norA基因中1155bp cDNA片段,将所得片段与pMD18-T载体连接,转化到感受态大肠杆菌JM109中,成功地筛选到阳性克隆,其质粒测序结果与文献报道一致。从阳性克隆中提取质粒,经Nde Ⅰ和XhoⅠ酶切,回收1155bp目的片段,定向克隆到pET28a(+)表达载体中,转化感受态大肠杆菌DH5a,提取质粒,再次转化到BL21(DE3)中,成功地筛选到阳性克隆。经IPTG诱导阳性菌,通过SDS-PAGE检测出norA基因的表达。

Abstract:根据文献设计1对PCR引物,寡聚核苷酸序列上游引物为5′-gCg,AAT,TCA,TCC,CAC,gTg,CCT,gC-3′,下游引物5′gAg,AAT,TCC,Tgg,ggA,ggg,ACC,TT-3′.经68℃退火及30轮循环的PCR程序后扩增出长度为1447 bp的DNA片段,将该片段从琼脂糖凝胶中回收,克隆到pMD18-T Vector后进行序列测定.序列分析结果表明,牦牛乳球蛋白基因5′调控区与文献报道的奶牛该基因同源性为97.65%,突出的碱基差异为牦牛乳球蛋白基因5′调控区发生了1个位点连续3个碱基的缺失突变和另一位点1个碱基的插入突变.该研究也为开展以牦牛乳球蛋白基因5′调控区为启动子的乳腺生物反应器研究准备了条件.

Abstract:在恒温7℃,10℃,14℃,18℃,22℃,26℃,29℃,32℃和35℃,对桔小实蝇在人工饲料上的发育和繁殖做了研究。结果表明,在7~32℃范围内桔小实蝇未成熟期各虫态的发育历期随温度的升高而缩短,卵的孵化率在26℃下最高;26℃下幼虫存活率最高,低温7℃、10℃以及14℃和高温35℃对卵、蛹的存活有明显的抑制作用,14℃蛹的死亡率达,与温度呈直线负相关;26℃繁殖率最高单雌产卵达682.2±23.62粒。

Abstract:建立了一种测定辣椒素和二氢辣椒素的反相高效液相色谱荧光法。样品中的辣椒素和二氢辣椒素用乙腈提取,以水和甲醇为流动相梯度洗脱,HP ODS Hypersil(5μm,250 mm×4 mm i.d)柱分离,用荧光检测器(λex=229 nm,λem=320 nm)测定。研究表明辣椒素和二氢辣椒在0.02~125.00μg/mL范围内,峰面积与待测物的质量浓度呈良好的线性关系,相关系数均为0.999 9,相对标准偏差在0.44%和2.74%之间,回收率为80.8%~99.9%。该方法操作简便、快速,适用范围广,重现性好,灵敏度高。

Abstract:采用高频电场激发氧灰化溴代卟啉分光光度法,测定了鄱阳湖野生藜蒿中铅的含量。在0.08 mol/LNaOH介质中,铅与溴代卟啉试剂形成1∶2橙黄色配合物,最大吸收波长在479 nm,表观摩尔吸光系数为2.2×105。铅量在0.04~0.48μg/mL内符合比耳定律,可用于鄱阳湖野生藜蒿中的铅的测定。测定结果表明,鄱阳湖区取样土壤已受到重金属铅的轻微污染,样品野生藜蒿中含有较低浓度的铅。

Abstract:从2003年11月至2004年10月,选取洞庭湖环湖丘岗区的4种生态系统——茶林、针阔混交林、阔叶林和竹林为样地,对其大型土壤动物群落的组成、多样性、垂直剖面分布及季节性变化等进行了比较研究。结果表明,被调查的4个土壤动物群落中动物类群数依次为竹林〉茶林〉阔叶林〉针阔混交林;个体数依次为茶林〉针阔混交林〉阔叶林〉竹林;多样性指数(H’)依次是竹林〉阔叶林〉茶林〉针阔混交林,与群落均匀度的变化显著相关,而与群落的优势度关系不密切;4个群落的DG指数是:竹林〉茶林〉阔叶林〉针阔混交林;通过对Jaccard指数分析结果表明,4种土壤动物群落组成具有较高的异质性,茶林和针阔混交林的相似性最高,茶林和阔叶林的相似性最低;4种群落的大型土壤动物类群及数量均具有表聚性,并随土层加深逐渐减少,且速度越来越快,尤其是随着季节变化波动性比较大。

Abstract:简要综述了近年来蕈菌交配型因子偏分离的研究概况。四极性蕈菌交配型A、B因子是非连锁的,双核菌丝体经原生质体单核化后2种交配型和担孢子4种交配型的比例应分别为1:1和1:1:1:1。但在实际的遗传分析中,许多蕈菌这2类交配型的比例并非总以1:1和1:1:1:1的形式出现。菌丝体经原生质体单核化后只找到1种交配型和担孢子只找到2种或3种交配型的现象并不罕见。香菇担孢子的4种交配型中,偏离孟德尔分离的担孢子以与亲本双核菌丝体相同的那2种交配型为多。对蕈菌交配型因子偏分离现象的系统研究报道极少,现有的一些试验结果认为:蕈菌交配型因子偏分离现象与单核体的再生能力和生长速度差异,与A、B交配型因子连锁的具致死效应的生育抑制因子的存在以及B交配型基因对自异核体分离的核存活的影响力不同有关。此外,可能导致交配型因子偏分离的因素还有交配型因子位点间或位点内重组,亲本交配型因子的异质状态以及细胞质基因组对核选择的细微影响等。
