Abstract:探讨了氯化支测定油菜籽 甙含量的方法中不同放置时间,不同温度,不同品种(系)和硫甙含量对测定结果的影响。结果表明:温度低于33℃时,放置4~16h均不影响硫甙含量测定。而35℃时,放置时间不能超过12h。低于33℃以下时,放置时间不能少于4h。但对于高硫甙品种,合适的温度和时间范围均变窄。且加入氯化式剂后,宜在20~27℃条件下显色,过高的温度会导致测定结果偏高。

Abstract:核盘菌(Sclerotiniasclerotiorum)引起包括油菜在内的多种经济作物的菌核病。为筛选抗菌核病的作物品种及探讨防治该病的其它途径,获得足够的子囊孢子是极其重要的技术环节。本研究对大量获得子囊盘的方法进行了改进。首先从158个单菌核分离物中筛选出了6个能够在高温(20℃)下直接形成子囊盘的核盘菌菌株。其次对形成菌核的温度条件、形成菌核的营养基质及菌核形成期等因于对子囊盘产生的影响进行了研究。结果表明所筛选的菌株在灭菌胡萝卜片上培育1个月即可产生大量的成熟菌核,这些菌核在20℃下保湿10-30d即可形成大量的子囊盘和子囊孢子。菌株Zxd-1的菌核最容易前发。试验还表明按这一程序获得的子囊孢子长出的菌丝能感染油菜,抗(耐)性品种(中油821)和感病品种(Westar)对接种体的反应表现出明显的差异。
