Detection of NBS-LRR type disease resistance gene in the resistance cultivars of tomato to Meloidogyne incognita

    Abstract:

    The resistance of 58 tomato Solanum lycopersicum materials to Meloidogyne incognita were studied by transplanting the tomatoes to disease soil in the pot. There were significant differences between 58 cultivars. The results showed that 1 cultivar was highly resistant,3 were resistant,3 were susceptible,51 were highly susceptible to Meloidogyne incognita. Degenerate oligonucleotide primers were designed according to the conserved NBS motifs among some disease-resistant genes. These primers were used for the amplification of the disease-resistant gene analog (RGA) from the genome of resistance cultivars by polymerase chain reaction (PCR), and about 500 bp-long PCR product was obtained.
