泥鳅二倍体与四倍体trim63a基因的克隆及表达分析

    试验以泥鳅(Misgurnus anguillicaudatus,Ma)为研究对象,采用PCR技术克隆获得trim63a基因(tripartite motif containing 63,又名肌肉环状指基因1)的cDNA部分序列,生物信息学分析结果显示,Matrim63a基因cDNA的编码序列(coding sequence,CDs)为1 035 bp,编码344个氨基酸,与硬骨鱼类相似度较高。采用荧光定量PCR技术检测人工繁殖获得的二倍体(D×D)和四倍体(T×T)胚胎不同发育时期Matrim63a的表达量以及泥鳅二倍体(D)和四倍体(T)成鱼不同组织中Matrim63a的表达量,基因表达分析结果显示,Matrim63a在泥鳅二倍体和四倍体成鱼肌肉中的相对表达量最高,其次是心脏;Matrim63a在泥鳅二倍体和四倍体胚胎的13个发育时期均有表达,但是不同倍性的胚胎表达差异较大。

    Abstract:

    Trim63(tripartite motif containing 63),also known as muscle RING finger protein 1 (MuRF1),plays an important role in the development and differentiation of the muscles in mammals.Partial cDNA sequence of the Misgurnus anguillicaudatus trim63a gene (Matrim63a) was cloned in this study.Sequence analysis showed that the fulllength Matrim63a CDs was 1 035 bp,encoding 344 amino acids,which shared high similarity with those of other teleosts.The Matrim63a mRNA expression in different tissues of the adults and during embryonic developmental stages in the diploid and tetraploid M. anguillicaudatus were examined by qPCR. Matrim63a showed highest expression in the muscle and followed by the heart.During embryonic development,Matrim63a was examined in all the 13 stages with different expression levels.
