      采用孢子萌发法和菌饼法评价β-蒎烯对意大利青霉菌的作用效果,分析β-蒎烯对病菌菌丝几丁质分布、菌丝活性氧代谢、排酸能力和麦角固醇含量的影响,并采用山梨醇、胆固醇或半胱氨酸作为保护剂探讨其对β-蒎烯抑制病菌孢子萌发的保护作用。结果显示,β-蒎烯对青霉菌表现出很强的抑菌活性,0.087~0.696 mg/mL的β-蒎烯能有效抑制孢子萌发与菌丝生长;β-蒎烯处理改变菌丝细胞壁几丁质分配,促进活性氧积累,降低菌丝排酸活性,显著降低菌丝中麦角固醇含量;添加麦角固醇替代物胆固醇、渗透调节剂山梨醇以及活性〖JP2〗氧清除剂半胱氨酸均能有效促进受0.348 mg/mL β-蒎烯抑制的青霉菌孢子萌发和芽管伸长。这些结果说明,β-蒎烯发挥对意大利青霉的抑菌作用与损伤真菌细胞壁结构、促进活性氧积累和抑制膜脂组分麦角固醇增加有密切关系。

      In this study,the effects of β-pinene on spore germination,tube elongation and mycelial growth of Penicillium italicum were assessed,and the effects of β-pinene on cell wall chitin distribution,active oxygen metabolism,acid excretion capacity and ergosterol contents of mycelia were analyzed.The effects of sorbitol,cholesterol and cysteine as protective agents on spore germination of P. italicum incubated with β-pinene were further evaluated.The results showed that β-pinene had strong antifungal activity against P. italicum in a dose dependent manner,as exhibited by spore germination and mycelium growth inhibition at the concentration ranging from 0.087 to 0.696 mg/mL.β-Pinene incubation also altered the distribution of chitin in cell wall,promoted the accumulation of reactive oxygen species (ROS),and significantly inhibited glucose-induced reduction in external pH of the mycelia.Addition of cholesterol as alternative of ergosterol,sorbitol as osmotic regulator,or cysteine as active oxygen scavenger could effectively promote the spore germination and tube elongation of the pathogen incubated with β-pinene at 0.348 mg/mL.These results revealed that the antifungal activity of β-pinene against P. italicum was related to the damage of cell wall structure,promotion of ROS accumulation and the inhibition of ergosterol increase in membrane.
