The filaments were embedded in glass bottom plate to fix filaments by using low melting point agarose. A real-time observation of microscopic culture system for Anabaena sp. strain PCC 7120 was prepared. Results showed that Anabaena sp. strain PCC 7120 grew well in glass bottom culture dish,and exhibited normal activity of cell division and differentiation. The continuous dynamics changes of cell division protein FtsZ and DNA double-stranded damage response of protein RecN during cell division process through this cultivation method were observed. It is indicated that continuous fixed-point observation of the same filament was successfully achieved.
