    为了提高香蕉线条病毒复合感染的诊断效率,以香蕉线条病毒OL(banana streak OL virus,BSOLV)、香蕉线条病毒GF(banana streak GF virus,BSGFV)和香蕉线条病毒IM(banana streak IM virus,BSIMV)为检测对象,分别根据BSOLV 和BSGFV的外壳蛋白基因(coat protein,CP)基因以及BSIMV的RNase H基因的保守序列设计特异性引物,以BSV多克隆抗血清捕捉BSV为模板,通过不同引物浓度,退火温度等条件的优化,建立了能够同时检测BSOLV,BSGFV,BSIMV的多重免疫捕捉PCR方法。研究结果表明,当BSOLV、BSGFV和BSIMV上下游引物浓度分别为0.5μM,退火温度为62.9℃时,建立的多重免疫捕捉PCR方法可以从1mg/mL 的香蕉粗汁液中分别检测出BSOLV、BSGFV和BSIMV。所建立的多重免疫捕捉PCR方法具有良好的特异性,与引起相似症状的黄瓜花叶病毒(cucumber mosaic virus, CMV)无交叉反应。这些研究结果表明,该多重免疫捕捉PCR方法具有较高的敏感性和特异性,可以应用于香蕉种苗以及田间香蕉样品中三种BSV的检测。

    Abstract:

    To improve the detection efficacy of banana streak virus co-infections, this study established multiplex immunocapture PCR for detection of co-infection of banana streak OL virus (BSOLV), banana streak GF virus (BSGFV) and banana streak IM virus (BSIMV). The specific primers were designed to identify the conserved sequences of the coat protein (CP) gene of BSOLV and BSGFV, and the RNase H gene of BSIMV. The BSVs were captured by BSV polyclonal antiserum as templates for a multiplex immunocapture PCR. After optimizing the reaction conditions including primer concentration and annealing temperature, a multiplex immunocapture PCR method was developed for simultaneous detection of BSOLV, BSGFV, and BSIMV. The results indicated that the multiplex immunocapture PCR technique could identify BSOLV, BSGFV, and BSIMV at each primer concentration of 0.5 μM from 1 mg/mL of banana sap at an annealing temperature of 62.9°C. The multiplex immunocapture PCR method was not able to detect the similar symptoms of banana caused by cucumber mosaic virus (CMV), and exhibited no cross-reactivity with CMV, indicating that the established multiplex immunocapture PCR method had good specificity. These results demonstrate that the multiplex immunocapture PCR method had high sensitivity and specificity for BSV, enabling its application in detecting the three BSVs in banana seedlings, as well as banana samples in the field.
