Effects of phthalic acid esters on nutritional quality and mechanisms of metabolic response in succulent stems of Zizania latifolia

    摘要:

    为探究邻苯二甲酸酯类化合物(phthalic acid esters,PAEs)胁迫下茭白肉质茎营养品质的变化机制,采用超高效液相色谱串联质谱结合非靶向代谢组学技术对不同质量分数PAEs处理下茭白肉质茎代谢机制进行系统解析。结果显示,高质量分数PAEs显著增加肉质茎粗纤维(46.15%)、可溶性蛋白(410%)和游离氨基酸(22.96%)含量,但降低白度、维生素C(50.11%)及可溶性固形物含量;中质量分数PAEs提高肉质茎总灰分(25.00%)和可溶性固形物含量(64.99%),而低质量分数PAEs下肉质茎粗纤维与还原糖含量最低。代谢组学鉴定差异代谢物689个。中质量分数PAEs诱导了227种代谢物变化,其中氨基酸类下调、脂类上调,显著影响了精氨酸和脯氨酸代谢通路;高质量分数PAEs引发267种代谢物变化,导致氨基酸类和苯丙素类上调,核苷酸类和有机酸下调。

    Abstract:

    The mechanisms of metabolic response in the succulent stems of Zizania latifolia under different concentrations of phthalic acid esters(PAEs) were systematically analyzed using ultra-high performance liquid chromatography-tandem mass spectrometry combined with untargeted metabolomics to study the mechanism underlying changes in nutritional quality of Zizania latifolia succulent stems under phthalate esters(PAEs) stress. The results showed that high concentrations of PAEs significantly increased the content of crude fiber(46.15%), soluble protein(410%), and free amino acids(22.96%) in the succulent stems, but decreased the whiteness, the content of vitamin C (50.11%), and the content of soluble solids. Medium concentrations of PAEs increased the content of total ash(16.67%) and water-soluble ash(11.48%) in the succulent stems, while the content of crude fiber and reducing sugars in the succulent stems were the lowest under low concentrations of PAEs. 689 differential metabolites were identified with untargeted metabolomics. Medium concentrations of PAEs induced changes in 227 metabolites, among which amino acids were down-regulated and lipids were up-regulated, significantly affecting the metabolic pathways of arginine and proline. High concentrations of PAEs triggered changes in 267 metabolites, leading to the up-regulation of amino acids and phenylpropanoids, and the down-regulation of nucleotides and organic acids. It will help to clarify the effects of PAEs pollution on the nutritional quality of crops, and provide a scientific basis for agricultural production and environmental protection to promote the sustainable development of agriculture and ensure food security.
