Morphological and molecular identification of awl nematode, Neodolichodorus sinensis associated with mangrove forest in Guangdong Province, China

    摘要:

    2021-2022年,分别在广东湛江红树林国家级自然保护区和广东镇海湾红树林国家湿地公园采集到2个新锥线虫种群样本。为探明该线虫的系统发育关系,利用形态学、分子生物学和系统发育的方法,将其鉴定为中华新锥线虫(Neodolichodorus sinensis),并获得中华新锥线虫湛江种群和镇海湾种群的18S rDNA和28S rDNA D2D3区序列数据。系统发育树构建结果显示:基于18S rDNA序列的系统发育树中中华新锥线虫与波斯湾新锥线虫(N. persiangulfus)聚为高度支持的单系分支;基于28S rDNA D2D3区序列的系统发育树中中华新锥线虫与波斯湾新锥线虫和白龙尾新锥线虫(N. bachlongviensis)聚为高度支持的单系分支。以上结果表明,中华新锥线虫与波斯湾新锥线虫互为姐妹种。

    Abstract:

    During 2021-2022, two awl nematode populations were collected from the Guangdong Zhanjiang Mangrove National Nature Reserve and the Guangdong Zhenhai Bay Mangrove National Wetland Park, respectively.To investigate the phylogenetic relationships among closely related species of Neodolichodorus, morphological, molecular and phylogenetic analyses were conducted.These analyses confirmed that both populations were identified as Neodolichodorus sinensis.The 18S rDNA and 28S rDNA D2D3 region sequences were obtained for the Zhanjiang and Zhenhai Bay populations of N.sinensis, and phylogenetic trees were constructed based on these sequences.The results showed that in the phylogenetic tree based on 18S rDNA sequences, N. sinensis formed a highly supported monophyletic clade with N. persiangulfus.In the phylogenetic tree based on the 28S rDNA D2D3 region sequences, N. sinensis clustered with N. persiangulfus and N.bachlongviensis in a highly supported monophyletic clade.This study clarifies the relationships between N. sinensis and its closely related species.This paper identifies N. sinensis and N. persiangulfus as sister cryptic species.
