Effects of inoculation quantity of Mesorhizobium huakuii on nodule formation and nitrogen fixation in Astragalus sinicus

    摘要:

    为探究接种华癸中慢生根瘤菌活菌数与紫云英植株结瘤固氮之间的关系,采用种子表面接种和植株根部接种2种方式,以液体菌剂为接种剂,通过无菌沙培系统,确定紫云英结瘤固氮所需接种根瘤菌的最低活菌数。结果显示,每粒种子表面接种根瘤菌数量达到102 CFU时,植株叶片开始由黄变绿,地上部鲜质量和株高较更低接种量处理显著增加;同时紫云英植株根部发育出具有固氮活性的根瘤,表现出良好的促生效果。对植株根部接种时,单株紫云英仅需满足101 CFU的接种量,即可开始结瘤固氮。无论选择哪种方式接种,单个紫云英植株接种根瘤菌数量达103 CFU时,其结瘤固氮作用、生物量和植株长势均明显优于低接种量组,继续增加接种量后,共生固氮表型趋于稳定。为充分保障根瘤菌接种剂在田间应用后的结瘤固氮作用,综合考虑菌剂施用的经济有效以及土壤条件的复杂性,确定紫云英单株接种103 CFU根瘤菌为华癸中慢生根瘤菌接种的最低活菌数。

    Abstract:

    Two inoculation methods including seed surface and root inoculations were used to study the effects of inoculation quantity of viable rhizobia Mesorhizobium huakuii on the nodule formation and nitrogen fixation in Astragalus sinicus. The minimum inoculation quantity of rhizobia required for the nodule formation and nitrogen fixation in A. sinicus was determined with a sterile sand-pot experiment system using liquid bacterial agents as inoculants. The results showed that the plant leaves started to turn from yellow to green, and the fresh weight and plant height aboveground significantly increased compared to that in treatments with lower inoculation quantity when the rhizobia concentration inoculated on the seed surface reached 102 CFU/seed. Meanwhile, nitrogen-fixing nodules developed on the roots of the A. sinicus plants, having a positive effect on promoting growth. When the plant root was inoculated, a single A. sinicus plant required only an inoculation quantity of 101 CFU/plant to initiate the nodule formation and nitrogen fixation. When the quantity of rhizobia inoculated on a single A. sinicus plant reached 103 CFU, its nodule formation and nitrogen fixation, biomass, and plant growth was significantly better than that in treatments with low inoculation quantity. Further increasing the inoculation quantity stabilized the symbiotic nitrogen fixation. 103 CFU/plant was determined to be the minimum inoculation quantity of M. huakuii rhizobia on A. sinicus to fully ensure the nodule formation and nitrogen fixation of rhizobia inoculants after application in the field, and take the economic effectiveness of inoculant application and the complexity of soil conditions into account.
