    为探究木薯普通花叶病毒(Cassava common mosaic virus,CsCMV)外壳蛋白CsCP在病毒侵染中是否通过与木薯谷氧还蛋白MeGRXC3互作协同调控氧化还原稳态,利用酵母双杂交(yeast two-hybrid,Y2H)、双分子荧光互补(bimolecular fluorescence complementation,BiFC)和亚细胞定位实验,对CsCP蛋白与MeGRXC3之间的相互作用进行研究。结果显示:MeGRXC3与CsCP在酵母细胞内存在相互作用;MeGRXC3与CsCP在烟草表皮细胞共表达后,在488 nm激发光下出现绿色荧光,证实CsCP与MeGRXC3在植物细胞内存在相互作用。亚细胞定位结果显示,CsCP与MeGRXC3在烟草表皮细胞中呈现共定位。以上结果表明,CsCP与MeGRXC3之间存在相互作用。

    Abstract:

    Cassava common mosaic virus (CsCMV) is the primary agent causing cassava mosaic disease.Its capsid protein (CP) plays a crucial role in virus-plant interactions during infection.Glutaredoxins (GRXs) are thiol oxidoreductases with low molecular weight, among which CC-type GRXs are key regulators of resistance to plant disease.Given that CP is a key pathogenicity factor of CsCMV involved in modulating host immune responses, this article investigated whether CsCP interacts with MeGRXC3 to synergistically regulate redox homeostasis and thereby modulate viral infection.The yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC), and subcellular localization assays were used to examine the interaction between CsCP and MeGRXC3.The results showed that there was an interaction between cassava MeGRXC3 and CsCP in yeast cells.Green fluorescence appeared at 488 nm after co-expressing MeGRXC3 and CsCP in Nicotiana benthamiana epidermal cells, confirming the interaction between CsCP and MeGRXC3 in plant cells.CsCP and MeGRXC3 exhibited co-localization in Nicotiana benthamiana epidermal cells.It is indicated that there is an interaction between CsCP and MeGRXC3.It will lay the groundwork for further elucidating the molecular mechanisms by which CP facilitates the infection of CsCMV.
